Common use

Clomid is an non-steroid anti-estrogen. Mechanism of its action consists in a specific binding with receptor for estrogen in ovary and hypophis. In patients with low estrogen levels in organism it causes estrogen effect and in ones with high estrogen levels it causes anti-estrogen effect. Its small doses increase secretion of gonadotropins (prolactin, Follicle stimulating hormone, FSH and interstitial cell-stimulating hormone), stimulates ovulation; in large doses slows secretion of gonadotropins. In the whole action of Clomid consists in stimulation of ovulation to occur and prepares the body for pregnancy. This medication may be used for male patients with infertility caused by low production of sperm.

Dosage and direction

Clomid is administered to stimulate ovulation 50 mg 1 time a day at bedtime, starting from 5th day of the menstrual cycle during 5 days (in the absence of the cycle – at any time). In case of no effect (ovulation does not occur within 30 days) increase the dose to 150 mg / day or lengthen the course to 10 days. Take exactly as prescribed, do not take the medication in larger quantities.

Precautions

Before to take Clomid notify your doctor if you have ever had any allergic reaction to clomiphene, liver diseases, mental depression, thrombophlebitis. This drug may cause vision problems, dizziness, or lightheadedness, be especially attentive if you need driving or operating machinery which requires high concentration of attention.

Contraindications

Hypersensitivity, liver or kidney failure, metrorrhagia of unknown etiology, ovarian cysts, neoplasms of genital organs, tumor or hypofunction of the hypophysis, endometriosis, ovarian failure together with hyperprolactinaemia, pregnancy.

Possible side effect

Together with allergy symptoms such as swelling of lips, tongue, or face or hives in some patients may appear the following conditions: ovarian enlargement presenting as abdominal or pelvic pain, flushing, nausea, vomiting, or diarrhea, blurred vision, headache, abnormal uterine bleeding.

Drug interaction

Inform your doctor and laboratory operators about all medications and herbal products you are taking if you are treated with Clomid.

Missed dose

If you missed a dose take it as soon as you remember, but not if it is almost time of the next intake by your schedule. If so skip the missed dose. Do not try to make up a missed dose by taking an extra one.

Overdose

If you suspect that you took too much of the medication seek for immediate medical attention.

Storage

Store Clomid in a dry place away from sunlight at room temperature between 15-30 C (59-86 F).

Common use

Progesterone is a type of female hormone (progestin). This medication is similar to the progesterone that your body naturally makes and is given to replace the hormone when your body is not making enough of it. Drug interaction

Common use

Danazol is a synthetic steroid produced from ethisterone. This medication suppresses synthesis of

gonadotrophic hormones luteinizing hormone (LH) and follicle-stimulating hormone (FSH) by pituitary gland in men and women. In women this drug inhibits activity of the ovaries, ovulation, leads to atrophy of the endometrium. The medication is used to treat to treat endometriosis (associated with sterility), fibrocystic breast disease, to prevent attacks of inherited angioedema, benign breast tumors (fibro-cystic breast), menorrhagia (abnormally heavy and prolonged menstrual period), premenstrual syndrome, primary premature puberty, gynecomastia.



Dosage and direction

The common recommended dose in adults is 200-800 mg a day divided in 2-4 intakes. Treatment starts on the first day of the menstrual cycle. To treat severe endometriosis the initial daily dose is 400 mg, later the dose may be increased to 800 mg by your physician. Treatment continues for six months. In children with premature puberty the dose is 100-400 mg.



Precautions

Cautiousness should be exercised in patients with diabetes mellitus, epilepsy, migraine. Use reliable and effective forms of contraception while you are on Danazol such as a condom or diaphragm with spermicide gel or inserts as hormonal forms of contraception may turn out non effective. Side effects

Danazol increases effects of anticoagulants of cumarine series and derivatives of indandione such as the risk of bleeding. Danazol decreases effects of insulin and oral hypoglycemic drugs by affecting metabolism of carbohydrates, increasing blood sugar levels Danazol inhibits metabolism of carbamazepine and increase its blood levels increases blood concentrations of cyclosporine and tacrolimus hereby increasing risk of nephrotoxicity. Danazol also increases serum levels of glucagon.

Common use

Premarin is a mixture of estrogen hormones which regulates development and functions of female reproductive system, formation of female secondary sex characteristics. On entering into the bloodstream it forms a complex with a specific receptor (in the fallopian tubes, cervix, excretory ducts of the mammary glands, vagina, urethra, breast, liver, hypothalamus, hypophisis), stimulates synthesis of DNA and proteins. Dosage and direction

Take this medication once daily as prescribed with a full glass of water with or without food. Premarin may be administered for a cyclic treatment which consists of 25 days on followed by 5 days. Do not take in larger amounts than prescribed. Individual dosage depends on your condition (for hypogonadism 0.3 mg or 0.625 mg daily, for preventing postmenopausal osteoporosis is 0.3 mg/day). Try your best to take it regularly to get the best benefit from it. Do not take with grapefruit juice, it increases levels of estrogen.



Precautions

Long-term treatment with Premarin may increase risk of breast cancer, endometrial hyperplasia, heart attack, or stroke so monitoring of the patient's state should be done on a regular basis. This medication can be harmful to an unborn baby, do not use it during pregnancy. Do not use Premarin to prevent hart disease or dementia. Hypersensitivity, pregnancy, breastfeeding, estrogen depending malignant tumors, undiagnosed genital or uterine bleeding, thrombophlebitis or thromboembolic disease in active phase (excluding treatment of breast and prostate cancers). Cautiousness should be exercised in patients with thrombophlebitis, thrombosis or thromboembolism (while taking estrogens in history); family hyperlipoproteinemia, pancreatitis, endometriosis, gallbladder disease in history (especially cholelithiasis), severe liver failure, jaundice (including one during pregnancy in history), hepatic porphyria, leiomyoma, hypercalcemia associated with bone metastases caused by breast cancer, underactive thyroid, porphyria, endometriosis, asthma.



Possible side effect

You may experience symptoms of allergy (hives, rash, swelling face and tongue) and also chest pain or heavy feeling, sudden numbness or weakness, abnormal vaginal bleeding, migraine headache, confusion, jaundice, swelling in hands, ankles, or feet. If such symptoms are severe stop taking this medication and notify your doctor at once.



Drug interaction

Estrogens stimulate the liver's ability to produce factors that promote the clotting of blood. Thus, concomitant administration of Premarin and warfarin (blood thinner) may cause decrease their effects. Barbiturates, Rifampin (Rifadin), phenytoin (Dilantin), carbamazepine (Tegretol), griseofulvin (Grifulvin), St. John's wort and primidone increase metabolism of estrogens. Erythromycin, ketoconazole (Nizoral), itraconazole (Sporanox), and ritonavir (Norvir) act oppositely.
